One of the most promising strategies aimed at reducing morbidity and preventing the risks of severe consequences of infection is the use of broad-spectrum antibodies that provide effective protection against infection with seasonal strains.</p> <p><bold>The aim</bold> of the study was to evaluate the protective activity of CR9114 antibodies of the IgG1 and IgA1 isotypes when administered systemically and locally against experimental influenza infection in mice.</p> <p><bold>Materials and methods.</bold> The recombinant antibodies CR9114 of IgG1 or IgA1 isotypes were administered intranasally to BALB/c mice at a dose of 100 or 20 μg 24 hours before infection with influenza virus A/California/07/09 (H1N1)pdm09 virus at a dose of 10 MLD<sub>50</sub> (prophylactic regimen) and/or 24 hours after infection (therapeutic regimen). Body weight dynamics were assessed and mortality was recorded in the animals for 14 days after infection.</p> <p><bold>Results. </bold>Intranasal administration of IgG1 or IgA1 isotype antibodies in the therapeutic-prophylactic regimen led to a decrease in viral load in the respiratory tract tissues of infected mice. At the same time, parenteral administration of IgG (but not IgA) also reduced the virus titer in the nasal passages (but not in the lungs) of mice. It was demonstrated that prophylactic administration of IgG1 or IgA1 antibodies provides complete protection against lethal influenza infection.</p> <p><bold>Conclusion. </bold>Intranasal prophylactic administration of human neutralizing antibodies CR9114 of IgG1 or IgA1 isotypes provides 100% survival of mice in lethal infection with influenza A/California/07/09 (H1N1)pdm09 virus. At the same time, Fc fragments of immunoglobulins of different isotypes, responsible for effector functions, appear to influence the degree of antiviral protection.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение. </bold>Грипп может вызывать заболевания разной степени тяжести, иногда приводящие к госпитализации или смерти. Одной из наиболее перспективных стратегий, направленных на снижение уровня заболеваемости и предотвращение рисков развития тяжёлых последствий инфекции, является применение антител широкого спектра действия, обеспечивающих эффективную защиту от заражения сезонными штаммами.</p> <p><bold>Цель</bold> исследования — оценка протективной активности антитела CR9114 изотипов IgG1 и IgA1 при системном и местном введении в отношении экспериментальной гриппозной инфекции у мышей.</p> <p><bold>Материалы и методы. </bold>Препараты полученных рекомбинантных антител CR9114 изотипов IgG1 или IgA1 интраназально вводили мышам линии BALB/c в дозе 100 или 20 мкг за 24 ч до заражения вирусом гриппа A/Калифорния/07/09 (H1N1)pdm09 в дозе 10 МЛД<sub>50 </sub>(профилактическая схема) и/или через 24 ч после инфицирования (терапевтическая схема). На протяжении 14 дней после заражения у животных оценивали динамику массы тела и проводили учёт летальности.</p> <p><bold>Результаты.</bold> Интраназальное введение антител изотипов IgG1 или IgA1 по лечебно-профилактической схеме приводило к снижению вирусной нагрузки в тканях респираторного тракта инфицированных мышей. При этом парентеральное введение IgG1 (но не IgA1) обеспечивало также снижение титра вируса в носовых ходах (но не в лёгких) мышей. Продемонстрировано, что профилактическое введение IgG1 или IgA1 обеспечивает полную защиту от летальной гриппозной инфекции.</p> <p><bold>Заключение.</bold> Интраназальное профилактическое введение человеческих нейтрализующих антител CR9114 изотипов IgG1 или IgA1 обеспечивает 100% выживаемость мышей при летальной инфекции вирусом гриппа A/Калифорния/07/09 (H1N1)pdm09.
