Genotyping and phylogenetic analysis of parvovirus B19 isolates isolated on the territories of Northwestern Federal District (NWFD) of Russia. Materials and methods. 61 blood sera and 30 oropharyngeal lavages obtained from patients with maculopapular rash from various territories of NWFD were studied for the presence of parvovirus B19 DNA (PVB19). DNA isolation and amplification was carried out by standard techniques. DNA segment including fragment of non-structural gene NS1 and region of structural gene VP1 (NS1-VP1u, 994 nucleotides) was sequenced, original sequences of oligonucleotide primers were selected for this purpose. Phylogenetic analysis was carried out online on the website http://www.phylogeny.fr. Data for tree construction was obtained from GenBank. Results. PVB19 DNA was detected in 45% ofsamples. PVB19 genome segment was sequenced in 8 samples. All the PVB19 isolates belong to a single cluster of 1A genotype. Isolate 57.12 from Komi Republic is similar to ISR-G strain isolated from Israel. Conclusion. Phylogenetic analysis showed a high degree of genetic similarity between PVB19 isolates circulating on the territories of NWFD, their membership in the most widespread genotype in the world. Local and import cases of parvovirus infection (PVI) were identified. The authors make a conclusion on the necessity to include PVI into the system of rubella and measles control.</p></abstract><trans-abstract xml:lang="ru"><p>Цель. Генотипирование и филогенетический анализ изолятов парвовируса В19, выделенных на территориях Северо-Западного Федерального Округа (СЗФО) России. Материалы и методы. На наличие ДНК парвовируса В19 (PV B19) исследовано 61 сыворотка крови и 30 ротоглоточных смывов, полученных от больных с макуло-папулезной сыпью с ряда территорий СЗФО. Выделение ДНК и амплификацию проводили по стандартным методикам. Секвенировали участок ДНК, включающий фрагмент неструктурного гена NS1 и область структурного гена VP1 (NS1 - VP1u, 994 нуклеотида), для чего были подобраны оригинальные последовательности олигонуклеотидных праймеров. Филогенетический анализ выполнен в режиме он-лайн на сайте http://www.phylogeny.fr. Данные для построения деревьев получены из GenBank. Результаты. ДНК PV B19 выявили в 45% образцов. Участок генома PV B19 был секвенирован в 8 образцах. Все изоляты PV B19 находятся в одном кластере генотипа 1А. Изолят 57.12 из республики Коми сходен со штаммом ISR-G, выделенным в Израиле. Заключение. Филогенетический анализ показал высокую степень генетической близости между изолятами PV B19, циркулирующими на территориях СЗФО, их принадлежность к наиболее распространенному в мире генотипу. Идентифицированы местные и завозной случаи парвовирусной инфекции (ПВИ).
