Development of a method of determination of anti-cytokine activity (ACA) of microorganisms, study of the prevalence and intensity of ACA to pro- and anti-inflammatory cytokines in pathogenic and opportunistic bacteria. Materials and methods. ACA was determined in 72 strains of microorganisms including members of the intestinal microflora and strains of pathogenic bacteria (salmonellae and gonococci). Study of the ability of supernatants and live cell cultures of microorgan- isms to induce changes in pro- (IFN-γ, TNF-α, IL-6) and anti-inflammatory cytokines (IL-4, IL-10) was performed by using co-incubation of exometabolites and live cell bacteria, fungi with recombinant cytokines. Results. A methodological approach allowing the determination of ACA, the prevalence of which among studied microorganisms was 50-62%, was developed. A decrease of cytokine concentration in the medium was registered in co-incubation of them with supernatants (in 56% of cases) and to a lesser degree - with live cell cultures (44%) of the studied bacteria and fungi. Expression of anti-cytokine activity was the most pronounced to TNF-α (Bifidobacterium spp. and Lactobacillus spp., Neisseria gonorrhoeae), IFN-γ (N.gonorrhoeae and Salmonella enterica) and IL-10 (Bifidobacterium spp. and Lactobacillus spp.). Conclusion. The data obtained expand the conception of modification of cytokine dynamic by pathogenic and opportunistic microorganisms, that can influence the course and outcome of an infectious process.</p></abstract><trans-abstract xml:lang="ru"><p>Цель . Разработать метод определения антицитокиновой активности (АЦА) микроорганизмов, изучить распространенность и выраженность АЦА в отношении про- и противоспалительных цитокинов у патогенных и условно патогенных бактерий. Материалы и методы. Определена АЦА у 72 штаммов микроорганизмов включая представителей кишечной микрофлоры и штаммы патогенных бактерий (сальмонеллы и гонококки). Изучение способности супернатантов и клеток живых культур микроорганизмов вызывать изменение концентрации про- (ИФН-γ, ФНО-α, ИЛ-6) и противовоспалительных цитокинов (ИЛ-4, ИЛ-10) проводили путем коинкубирования экзометаболитов и живых клеток бактерий, грибов с рекомбинантными цитокинами. Результаты. Разработан методический подход, позволивший определить АЦА, распространенность которой среди исследуемых микроорганизмов составляла 50-62%. Снижение концентрации цитокинов в среде отмечено при коинкубировании их с супернатантами (в 56% случаев) и в меньшей степени - с клетками живых культур (44%) исследуемых бактерий и грибов. Экспрессия антицитокиновой активности была максимально выражена в отношении ФНО-α (Bifidobacterium spp. и Lactobacillus spp., Neisseria gonorrhoeae), ИФН-γ (N.gonorrhoeae и Salmonella enterica) и ИЛ-10 (Bifidobacterium spp. и Lactobacillus spp.). Заключение.
